A peptic ulcer is a disease that affects the stomach lining caused by Helicobacter pylori. The present research aimed to formulate and evaluate mucoadhesive microsphere of ofloxacin for effective safe treatment ulcers. microspheres were developed using natural polymers like Guar gum, Sodium alginate, Chitosan emulsification cross-linking method. total 8 batch formulations prepared 23 full factorial designs. evaluated various parameters such as % yield, entrapment efficiency, drug release study, in vitro wash-off test, optimization was done design expert which further subjected statistical analysis kinetic mechanism, compatibility studies performed FTIR DSC. Using maximum possible formulation, optimized formulation found be F5 terms better longer time with properties on use guar sodium chitosan ratio 1:1:2.
